Yes, the button is immediately detected. Please note, however, each controller reports the button states at certain intervals. For PS4 controller is each 4ms, for XB1 controllers is each 8ms.
If you are using a PS4 controller, you can change this interval to 1ms by setting the Titan Two input polling rate to 1000Hz. Unfortunately, the XB1 controller still reports the buttons states at 8ms even with the input polling rate at 1000Hz.

did i get it right ?
There is no delay difference between normal motion and macro.
Another question: would there be an extra delay if I play with the Xbox one controller on the playstation 4 console

Yes, despite the Titan Two be able to update the PS4 at 1000Hz, the updates coming from the XB1 controller will still at 125Hz (8ms). However, any script or macro running on the Titan Two will still be able to update the PS4 at 1000Hz.
